چکیده

W e describe a 16-year-old boy with juvenile metachromatic leukodystrophy who was treated with bone marrow transplantation. Follow-up over 8 years showed no increase in symptoms, no progression of neurological signs, and no neuropsychological deterioration. We conclude that bone marrow transplantation may increase brain levels of arylsulfatase A enough to prevent deterioration in patients with juvenile metachromatic leukodystrophy. Serial MR after bone marrow transplantation in two patients with metachromatic leukodystrophy.

Two children with metachromatic leukodystrophy underwent bone marrow transplantation. In both patients MR subsequently showed, first, white matter changes, then later, lack of change as the patients stabilized clinically.

Improvement of white matter changes on neuroimaging modalities after stem cell transplant in metachromatic leukodystrophy.

IMPORTANCE We sought to illustrate improvement of cerebral white matter changes in metachromatic leukodystrophy after treatment with hematopoietic stem cell transplant (HSCT). OBSERVATIONS We conducted serial magnetic resonance imaging (MRI) and proton magnetic resonance spectroscopy (1H-MRS) as standard follow-up after HSCT with cord blood in 1 patient with juvenile metachromatic leukodystro...

Infantile metachromatic leukodystrophy in an 18 month old girl.

Metachromatic leukodystrophy is a rarely occurring neurodegenerative metabolic disorder with an incidence of 1-9 individuals out of 1,000,000. We present a similar case in an eighteen month old child which was extremely challenging to diagnose. Clinical symptoms suggested motor regression and developmental delay which gave rise to suspicion of a neurodegenerative disorder. An MRI scan of the br...

Juvenile metachromatic leukodystrophy in a boy with epilepsy.

M leukodystrophy (MLD) is an autosomal recessive disorder, caused by deficiency of arylsulfatase A (cerebroside sulfatase A, ASA), which results in accumulation of sulfatides, mainly in the brain and peripheral nerves. The estimated frequency of MLD from previous reports is 1 in 40,000 cases. The deficiency in ASA activity is caused by mutations in the ASA gene. The deficiency is systemic and a...

Long-term outcomes after allogeneic hematopoietic stem cell transplantation for metachromatic leukodystrophy: the largest single-institution cohort report

BACKGROUND Metachromatic Leukodystrophy (MLD) is a rare, fatal demyelinating disorder with limited treatment options. Published outcomes after hematopoietic stem cell transplantation (HSCT) are scant and mixed. We report survival and function following HSCT for a large, single-center MLD cohort. METHODS Transplant-related data, survival and serial measures (brain MRI, nerve conduction velocit...
